APSTRAKT
Cilj. Prolinom bogati polipeptidi, galarmin i analozi, su
novi neuropeptidi koje je u neurosekretornim granulama
hipotalamusa otkrio i izolovao prof. A. Galoyan sa sarad-
nicima. Pokazano je da galarmin i analozi imaju veliku
biološku aktivnost uključujući snažnu in vivo imunomod-
ulatornu i antibakterijsku aktivnost protiv velikog broja
dobro poznatih patogena. Cilj ove studije je bio da se ispita
direktna antibakterijska aktivnost galarmina i analoga pro-
tiv različitih Gram-pozitivnih i Gram-negativnih bakterija
uključujući MRSA sojeve in vitro.
Metod. Direktna antibakterijska aktivnost galarmina i
analoga je odredjivana esejom inhibicije rasta u tečnosti
bakterija protiv različitih sojeva. Bakterije su gajene u Lu-
rija-Bertani standardnom tečnom medijumu tokom 2-3h
na 37°C, 250 o/min. Minimalne inhibitorne koncentracije
(MIC) za galarmin i analoge su utvrđene merenjem apsor-
pcije (optičke gustine - OG) udubljena u ploči, na 630 nm,
pomoću čitača ploča (Asys UVM 340). Statistička analiza
svih podataka izvršena je Studentovim i Fišerovim testom,
pri verovatnoći od p≤0.05.
ABSTRACT
Objective. Proline-rich polypeptides, galarmin and ana-
logues, are new neuropeptides discovered and isolated from
neurosecretory granules of hypothalamus by Prof. A. Galoyan
and coworkers. It was shown that galarmin and analogues
have large biological activity including strong immunomodu-
latory and antibacterial activity in vivo against a number of
well knows pathogen. The purpose of this study was to inves-
tigate the direct antibacterial activity in vitro of galarmin and
analogues against different Gram-positive and Gram-negative
bacteria, including MRSA strains.
Method. Direct antibacterial activity of galarmin and its
analogues was monitored by a liquid growth inhibition assay
against different bacterial strains, Bacteria were cultivated in
Luria-Bertani standard liquid medium during 2-3h at 37°C, 250
rpm. The minimal inhibitory concentrations (MICs) of galar-
min and analogues were determined by measuring the absor-
bance (optical density - OD) of the plate’s wells at 630 nm by
plate automatic reader (Asys UVM 340). Statistical analysis of
all data was performed by Student’s and Fishers’ methods, at
probability of p≤0.05.
